Assessing the immunomodulatory potential of high‐molecular‐weight extracts from mushrooms; an assay based on THP‐1 macrophages

Abstract: An assay was developed suitable to distinguish the immunomodulatory properties of food compounds in a reproducible manner. It was evaluated using eight mushroom species by measuring the secretion of relevant cytokines TNF-α, IL-1β, IL-6 and IL-10.

“…[72,73] LPS-stimulated THP-1 cells are commonly used to evaluate the immunomodulatory activity of natural products. [74][75][76] LPS is present in the outer membrane of Gram-negative bacteria and is a key pathogen-associated molecular pattern that interacts with TLR4 receptors to trigger the release of cytokines by monocytes and macrophages. [77][78][79] We observed that LPS-stimulated THP-1 cells produced TNF-α and IL-12, and that FE extracts at all tested concentrations significantly reduced the levels of these cytokines.…”

“…The pattern recognition receptors activate the innate immunity after pathogen recognition, while Tolllike receptors activate the pathways coordinating with the innate immunity and trigger the immunity response [114]. Velde et al studied the immunomodulatory potential of the Agaricus subrufescens and Coprinus comatus species of mushroom using the THP-1 cells [116]. Various other researchers also evaluated the upregulation of genes using the extracts of Ganoderma lucidum, L. edodes, Agaricus bisporus, and A. subrufescens [117][118][119][120].…”
