Newcastle disease (ND) is a highly contagious viral disease. This study was conducted to determine the antibody titer in Japanese quails following vaccination against ND and feed-supplementation with vitamin-C, probiotics and antibiotic-growth-promoters. Forty Japanese quails were equally grouped in four groups (A1 to A4) and were vaccinated by LaSota, B1, F and VG/GA strains, respectively, at the age of 10 days. Another forty Japenese quails were equally grouped in four groups (B1 to B4), were vaccinated with the vaccines of the previous groups that showed a higher effect, and were fed with mesh-feed (control-B1), supplemented with vitamin-C (B2), probiotics (B3), and antibiotic growth promoter (B4). Serum samples were obtained on 2, 4, 6, 8, 10 and 12 weeks of age and were subjected to a hemagglutination inhibition test. The mean antibody titer (Log2) had no significant difference between groups A1-A4. The highest titers were recorded in A4 (6.20±0.37) and A3 (6.00±0.71). The mean titer was significantly different among the groups at 4 weeks, but insignificant in the other weeks of the supplemented groups. A higher mean titer was found between 8 and 10 weeks in group B2 (7.40±0.60) and B3 (7.20±0.58). The relative weights (gm/kg) of the spleen, thymus and bursa were significantly higher in group B3 (1.947±0.109, 5.951±0.312 and 2.624±0.083, respectively). They had no significant correlation with antibody titer at 12 weeks. In conclusion, vitamin-C and probiotic supplemented feeds can boost up the antibody titer against ND in Japanese quail when given during VG/GA strain vaccination.

Background Due to the more stability and a better homogenecity in immune response, the use of thermoresistant vaccines in different chicken types has been increased. Objective This study aimed to evaluate the efficacy of a newly developed Newcastle disease vaccine (ND.TR.IR) originating from I‐2 strain in specific pathogen‐free (SPF) and native and broiler chickens. Methods Following determination of pathogenicity indices on the candidate seed, three efficacy examinations were conducted. In the first experiment, 120 1‐day‐old SPF chickens were randomly allocated to six groups and either vaccinated with ND.TR.IR via eye drop at 1, 7, and 21 days of age (V1, V7, and V21), or considered as non‐vaccinated control groups (C1, C7, and C21). At 20th post‐vaccination day, sera hemagglutination inhibition (HI) antibody titres against ND virus (NDV) were measured and then the chickens were challenged by virulent NDV (vNDV). In the second and third experiments, the efficacy of ND.TR.IR vaccine was compared to routine vaccination program (B1 and LaSota) in native and broiler chickens that were vaccinated at 10 and 20 days of age, respectively. The HI antibody titres were measured on 10, 20, 30, and 40 days of age, and also challenge efficacy test with vNDV was conducted on 30 days of age. Results The studied virus, as a vaccinal seed, complied with the pathogenicity indices of avirulent NDV and molecular identity of I‐2 strain. In the efficacy evaluation trials, the vaccinated chickens had higher HI antibody titres against NDV compared with their corresponding control chickens (p < 0.05). Results of the challenge tests indicated 95% and 100% protection against vNDV in native, SPF, and broiler‐vaccinated chickens, respectively. Conclusions The present findings indicated that administration of ND.TR.IR induced appropriate HI antibody titres against NDV in SPF, native, and broiler chickens associated with good protection in efficacy test.

AbstractThe administration of antibiotics to day old chicks as a means of prevention or treatment of suspected hatchery or farm-borne infections is common, especially in developing countries. This practice could contribute to a poor immune response following Newcastle disease (ND)-LaSota vaccinations, in addition to the sluggish growth in broiler chickens. This study was aimed at determining: the antibody titre to ND-LaSota vaccine, live weight, weight gain and feed conversion efficiency (FCE) of broiler chicken exposed early to gentamicin and doxycycline. One hundred, day-old broiler chicks were randomly assigned to four groups (n = 25). Group 1 served as a control, while groups 2 and 4 received gentamycin and doxycycline, respectively. The chicks in group 3 were treated with a combination of gentamicin and doxycycline (1 : 1). All drugs were administered via the drinking water from the 2nd to the 6th day of the chicks’ life. On day 18, the birds received ND-LaSota vaccine intraocularly. At weekly intervals, the post-vaccination antibody titre, live weight and weight gain were determined. The feed conversion efficiency (FCE) of the different groups was calculated at the end of the experiments. The results showed that the NDV antibody titre of the antibiotic-treated groups did not differ significantly (P < 0.05) from that of the control. However, there was a significant (P < 0.05) increase in the live weight, weight gain and FCE of the control birds when compared to the antibiotic-treated groups.
