Association of an isozyme locus and strawbreaker foot rot resistance derived from Aegilops ventricosa in wheat

Abstract: Thirty lines from a cross between VPM/ Moisson 421 and Selection 101 were used in the study to determine whether strawbreaker foot rot resistance derived from Aegilops ventricosa was associated with an allele for endopeptidase. The progeny examined for foot rot lesions represented F2 derived F5 lines and enzyme assays were done on F6 seedlings. The results indicate that the wheat and 'VPM/Moisson 421' endopeptidase alleles are distinctly different. The endopeptidase allele frequencies of 30 lines were compared… Show more

“…Close association between Pch1 and a codominant endopeptidase marker allele, Ep-D1b, was demonstrated by McMillin et al (1986) and subsequently used to monitor introgression of eyespotresistance (Pch1) from VPM-1 to elite lines. Improvements in the endopeptidase assay have recently been published and this assay was found to be completely accurate in predicting resistance (Santra et al 2006).…”

“…(Maia 1967) and is located on the distal end of chromosome 7D (Doussinault et al 1983;Worland et al 1988). According to McMillin et al (1986), there is a close association between the Pch1 resistance gene and EP-D1b, an endopeptidase isozyme allele from Ae. ventricosa chromosome 7DL.…”

“…jic-5A loci) and negative (Ozon and Patras, eyespot susceptible) control varieties. Pch1, Pch2 and Q.Pch.jic-5A loci identification was made using eight verified markers (Table 1) according to McMillin et al (1986); Groenewald et al (2003); Chapman et al (2008) and Burt et al (2011). Based on the marker analysis, we selected five groups of breeding lines, carrying: (1) the Pch1 gene alone: one line; (2) the Pch2 gene alone: four lines; (3) the Q.Pch.jic-5A alone: one line; and (4) Pch1 + Q.Pch.jic-5A: three lines.…”

“…"Marne" (Maia 1967 (Doussinault et al 1983). The transferred sequence with the Pch1 gene has been mapped in chromosome arm 7DL (Jahier et al 1978;McMillin et al 1986;Worland et al 1988). "VPM1" line was a base for breeders to create new resistance cultivars like "Rendezvous" (used in the presented work as a resistance standard).…”

“…An identification of the Pch1 locus is provided by closely linked endopeptidase gene locus EpD1b (McMillin et al 1986;Worland et al 1988;Groenewald et al 2003). Endopeptidase-1 (Ep-1) in wheat is controlled by 3 loci: EpA1, EpB1 and EpD1, located in homoeologous chromosomes 7AL, 7BL and 7DL, respectively (Hart and Langston 1977;Tuleen 1977, Koebner et al 1988;Santra et al 2006).…”
