Association of protein tyrosine phosphatase non-receptor type 22 gene functional variant C1858T, HLA-DQ/DR genotypes and autoantibodies with susceptibility to type-1 diabetes mellitus in Kuwaiti Arabs

Abstract: The incidence of type-1 Diabetes Mellitus (T1DM) has increased steadily in Kuwait during recent years and it is now considered amongst the high-incidence countries. An interaction between susceptibility genes, immune system mediators and environmental factors predispose susceptible individuals to T1DM. We have determined the prevalence of protein tyrosine phosphatase non-receptor type 22 (PTPN22) gene functional variant (C1858T; R620W, rs2476601), HLA-DQ and DR alleles and three autoantibodies in Kuwaiti child… Show more

“…The hallmark of HLA susceptibility is, however, considered from the haplotype point of view [31, 33, 34, 43, 44, 5155]. Reported Arab T1D HLA studies were tabulated in Table 3 [49, 50, 5560], which highlights the dearth need for systematic HLA studies in these populations. A recent meta-analysis published in 2015 analyzed 23,333 articles, of which only 30 were based on an Arab population.…”

“…There is a dearth amount of information about autoantibodies in type 1 diabetes in Arab populations (Table 4). Majority of the present studies are case control and investigated the presence of only GADA and IA2 [49, 50, 69–73]. There are interesting studies correlating gene polymorphisms and the ICA, IA2, and GADA to the susceptibility to type 1 diabetes in the Kuwaiti population [49, 50].…”

“…The genetic background is known to influence susceptibility to diabetes development, insulin resistance, and the efficacy of glucose-lowering drugs [17]. Accumulated evidence suggests that ABCA1 contributes to secreting insulin in pancreatic β-cells and amyloid beta for- mation [18][19][20].…”

“…A previously described method was used to isolate total genomic DNA [17]. The genotypes of a functional variant +1858C→T (rs2476601) in the PTPN22 gene were identified by polymerase chain reaction-restriction enzyme fragment length polymorphism (PCR-RFLP) method [18,19]. In order to amplify a 218 bp DNA fragment, the primers: Forward primer: 5'-ACTGATAATGTTGCTTCAACGG-3' and reverse primer:5'-TCACCAGCTTCCTCAACCAC-3' were used.…”
