Association of RGA-SSCP markers with resistance to downy mildew and anthracnose in grapevines

Abstract: ABSTRACT. Downy mildew (Plasmopara viticola) and anthracnose (Sphaceloma ampelinum) are two major diseases that severely affect most grapevine (Vitis vinifera) cultivars grown commercially in Thailand. Progress of conventional breeding programs of grapevine for improved resistance to these diseases can be speeded up by selection of molecular markers associated with resistance traits. We evaluated the association between 13 resistance gene analog (RGA)-single-strand conformation polymorphism (SSCP) markers with… Show more

“…The symptoms were assessed in variable times of incubation. The results obtained by these assays were consistent and were applied for pathogenicity analysis (Tharapreuksapong et al, 2009; Santos et al, 2018a), germplasm characterization (Kono et al, 2012) and genetic analysis (Tantasawat et al, 2012; Poolsawat et al, 2013).…”

“…However, no information was found about the use of the SCAR in routine breeding programs to assist the introgression of the resistant gene into V. vinifera germplasm. Recently, also RGA-SSCP markers have been found associated with AN resistance (Tantasawat et al, 2012).…”

“…Recently, we found three resistance gene analog (RGA)-single-strand conformation polymorphism (SSCP) markers that were associated with downy mildew and anthracnose resistance in crosses using 'NY65.0550.04' as a resistance source. These markers could be used to accelerate the selection process for improved downy mildew and anthracnose resistance, particularly in the 'Carolina Black Rose x NY65.0550.04' cross in future breeding programs (Tantasawat et al, 2012).…”

“…The SCAR marker OPB 151247 was linked to the anthracnose resistance locus in grape (Kim et al, 2008 ). Three resistance gene analog‐single‐strand conformation polymorphism (RGA–SSCP) markers were subsequently found with significant correlation with anthracnose resistance using the interspecific hybrid NY65.0550.04 as a resistance source (Tantasawat et al, 2012 ). The markers from different resistance sources seem to have limitations of application to some extent.…”
