Association of Tartrate-Resistant Acid Phosphatase-Expressed Macrophages and Metastatic Breast Cancer Progression

Abstract: Infiltrating neutrophils, lymphocytes, macrophages, and cytokines constitute a state of chronic inflammation within the tumor microenvironment. Tartrate-resistant acid phosphatase 5a (TRACP5a) protein, a novel product of activated macrophage, is postulated to be a biomarker for systemic inflammatory burden in states of chronic inflammation. We aimed to investigate the clinical significance of TRACP5a expression in tumor-infiltrating macrophages and serum TRACP5a in patients with metastatic breast cancer (BC).W… Show more

“…ACP5 encodes tartrate-resistant acid phosphatase (TRAP) that is a metalloenzyme expressed in activated osteoclasts and macrophages, which has recently gained traction as a driving factor for metastasis and is significantly associated with cancer progression (46, 47). Overexpression of TRAP increases anchorage-independent and anchorage-dependent cell growth and proliferation and promotes cell migration and invasion (47).…”

“…TRACP-5a is also reported to be a biomarker for many benign or malignant diseases [ 6 – 10 ]. Chen et al reported that the serum TRACP-5a level lower than 12.4 μg/L had better survival for metastatic breast cancer [ 7 ]. The serum TRACP-5a level also elevated significantly in active sarcoidosis compared with control group (11.66 versus 8.04 μg/L) [ 8 ].…”

“…The convincing link between chronic inflammation, breast cancer risk, and acid phosphatase activity shown by other authors 41 was the basis for our next step in elucidating the role of the acid phosphatase (AcP) enzyme in a comparative analysis of tissue exposure and carcinogenesis models in animals. The results of this experimental series are shown in Table 2 .…”

“…Recently, the role of biomarkers of specific lysosomal enzyme complexes has been reported in a number of papers. They described the role of AAP in solid and hematologic tumors 37 , 38 , LAP in the progression and metastasis of several cancers 39 , 40 , AcP as a link between chronic inflammation and breast cancer 41 , NAG in luminal breast epithelial cells 42 , and β-GD in the proliferation of breast cancer cells 43 .…”
