Associations between the Expression of Epigenetically Regulated Genes and the Expression of DNMTs and MBDs in Systemic Lupus Erythematosus

Abstract: ObjectivesWe determined the expression of ITGAL, PRF1, KIR2DL4, CD70, and CD40LG in patients with SLE and performed correlations with the global DNA methylation status and the levels of three DNA methylation enzymes and two methyl CpG-binding domain (MBD) proteins.Patients and MethodsCD4+ T cells were isolated from 35 SLE patients and 30 healthy controls. DNA deoxymethylcytosine content was measured by an enzyme-linked immunosorbent assay (ELISA). Transcript levels of ITGAL, PRF1, KIR2DL4, CD70, CD40LG, DNMT1,… Show more

“…Together, DNA hypomethylation may account for aberrant gene expression in SLE. This could be the result of defective ERK activation and subsequent DNMT1 downexpression and also overexpression of DNA demethylation enzymes in SLE T cells [31][32][33][34][35].…”

“…Recently, Balada and coworkers determined the expression levels of ITGAL , PRF1 , KIR2DL4 , TNFSF7 ( CD70 ), and CD40LG genes in CD4+ T cells of patients with SLE and performed correlations with the global DNA methylation status and the levels of DNMT and MBD proteins. SLE patients had signifi cantly elevated transcript levels of ITGAL , PRF1 , and TNFSF7 ( CD70 ), and those levels correlated with global DNA hypomethylation as well as with the expression of most of the DNA methylation-related genes, reinforcing the hypothesis of an epigenetic deregulated network in SLE (Balada et al 2012 ).…”

“…Most loci on the X chromosome of females are subject to X-chromosome inactivation (only one allele from each pair of alleles is expressed) 15. However, a recent report raised the possibility that some loci on the X chromosome of females can escape X-chromosome inactivation 16. For example, females had higher levels of CD40LG than males16 and therefore the genetic risk to a male hemizygote (A/-) may not be equivalent to the genetic risk to a female homozygote (A/A), which requires alternate hypotheses for testing association on the X-chromosome loci 11,17.…”
