Sex-determining systems may evolve rapidly and contribute to lineage diversification. In fact, recent work has suggested an integral role of sex chromosome evolution in models of speciation. We use quantitative trait loci analysis of restriction site-associated DNA -tag single nucleotide polymorphisms to identify multiple loci responsible for sex determination and reproductively adaptive color phenotypes in Lake Malawi cichlids. We detect a complex epistatic sex system consisting of a major female heterogametic ZW locus on chromosome 5, two separate male heterogametic XY loci on chromosome 7, and two additional interacting loci on chromosomes 3 and 20. Our data support the known chromosomal linkage between orange blotch color and ZW, as well as novel genetic associations between male blue nuptial color and two sex determining regions (an XY and ZW locus). These results provide further empirical evidence for a complex antagonistic sex-color system in this species flock and suggest a possible role for, and effect of, polygenic sex-determining systems in rapid evolutionary diversification.

We used molecular approaches to study the status of speciation in coral reef fishes known as hamlets (Serranidae: Hypoplectrus). Several hamlet morphospecies coexist on Caribbean reefs, and mate assortatively with respect to their strikingly distinct colour patterns. We provide evidence that, genetically, the hamlets display characteristics common in species flocks on land and in freshwaters. Substitutions within two mitochondrial DNA (mtDNA) protein-coding genes place hamlets within a monophyletic group relative to members of two related genera (Serranus and Diplectrum), and establish that the hamlet radiation must have been very recent. mtDNA distances separating hamlet morphospecies were slight (0.6 ± 0.04%), yielding a coalescent estimate for the age of the hamlet flock of approximately 430 000 years. Morphospecies did not sort into distinct mtDNA haplotype phylogroups, and alíeles at five hypervariable microsatellite loci were shared broadly across species boundaries. None the less, molecular variation was not distributed at random. Analyses of mtDNA haplotype frequencies and nested clades in haplotype networks revealed significant genetic differences between geographical regions and among colour morphospecies. We also observed significant microsatellite differentiation between geographical regions and in Puerto Rico, among colour morphospecies; the latter providing evidence for reproductive isolation between colour morphospecies at this locale. In our Panama collection, however, colour morphospecies were mostly genetically indistinguishable. This mosaic pattern of DNA differentiation implies a complex interaction between population history, mating behaviour and geography and suggests that porous boundaries separate species in this flock of brilliantly coloured coral reef fishes.

Descriptions of five new species of <i>Metriaclima</i> (Teleostei: Cichlidae) from Lake Malaŵi, Africa

Lake Malaŵi is known for its endemic haplochromine species flock, most notably the rock-dwelling cichlids known as mbuna. One of the larger genera of mbuna is Metriaclima, a group consisting of 31 described species (including the five described herein) and approximately 45 recognized unique populations. Metriaclima is diagnosed by its feeding behavior and several morphological characteristics including the angle of the vomer and the presence of bicuspid teeth in the outer row of both the upper and lower jaws. Metriaclima zebra, the type species for the genus, was described based on a single specimen. While the collection location of this holotype is not known, based on the travel records of its collector, it is likely that the specimen originated from Likoma Island. The holotype was therefore compared to specimens from several localities around this island and was found to be morphologically indistinguishable from some of these. This study includes the morphological analysis of 496 specimens of Metriaclima belonging to 31 collections from Lake Malaŵi. Morphometric differences were analyzed and the relationships among several distinguishable populations of Metriaclima zebra were investigated. Our study further resulted in the description of the following five new species belonging to the M. zebra species com-plex: M. pambazuko, M. lundoense, M. midomo, M. tarakiki, and M. nigrodorsalis. These species were distinguished and described based on color patterns, morphometric, meristic, and ecological differences. These new species were compared with and distinguished from nearby populations of Metriaclima having similar pigmentation patterns and/or similar eco-logical niches. An artificial dichotomous key to the described species of Metriaclima is presented.
