Australian and Pacific contributions to the genetic diversity of Norfolk Island feral chickens

Abstract: BackgroundNorfolk Island has a population of feral chickens which could be the result of domestic stock introduced onto the island by British settlers in 1788. However, there is ongoing debate about their origins because multiple human arrivals to the island may have brought chickens with them. Here we investigate the genetic origins of these feral chickens by sequencing their mitochondrial control region. We infer their phylogenetic relationships using a large dataset of novel sequences from Australian mainla… Show more

“…Haplogroups A, B, and CD of mtDNA D-loop sequences are mainly distributed in chickens from East Asia, Southeast Asia, and the Pacific islands 4 , 5 , 28 . The haplotypes of haplogroups A and/or CD that were observed in 15 populations of indigenous chickens and red junglefowl in Thailand were similar or closely related to the representative haplotypes in South China, Vietnam, Laos, Sri Lanka, and Japan 5 .…”

“…DNA was extracted using QIAamp DNA Investigator Kit (Qiagen, Hilden, Germany). The mtDNA CR was chosen as the target as it is highly polymorphic and phylogeographically informative [3,14,20,33,41]. The target region of mitochondrial hypervariable region 1 was amplified using the following primer set: 5 0 -AGGACTACGGCTTGAAAAGC-3 0 and 5 0 -ATGTGCCTGACCGAGGAACCAG-3 0 .…”
