Autohydrolysis of a partially cyclized m/u-carrageenan and structural elucidation of the oligosaccharides by chemical analysis, NMR spectroscopy and UV-MALDI mass spectrometry

Abstract: Dedicated to Prof. Rosa M. de Lederkremer on occasion of her 70 th anniversaryAbstract A partially cyclized mu/nu-carrageenan from Gigartina skottsbergii was submitted to autohydrolysis and the resulting mixture of oligosaccharides was fractionated using sequential dialysis through tubings of molecular weight cutoff 12000, 3500 and 1000. In this way four fractions were separated and characterized. Structural analysis of the major fraction, obtained from the solution inside the dialysis tubing of molecular weig… Show more

“…The mild acidic conditions generated in the media during the hydrothermal process could lead to the fragmentation of glycosidic linkages. A similar trend was reported for the autohydrolysis of a partially cyclized mu/nu-carrageenan from S. skottsbergii caused after 11 h at 60 °C, from the initial 198 kDa carrageenan to fractions lower than 1.9 kDa [ 28 ].…”

“…The H-type fractions, obtained by autohydrolysis, display similar inhibitory activities as the corresponding unfractionated material ( Figure 4A ). C2-selective desulfation of fucopyranose (Fuc) and degradation happen during the process of autohydrolysis ( Ciancia et al, 2005 ; Pomin et al, 2005 ; Anastyuk et al, 2012 ; Rodriguez-Jasso et al, 2013 ; Menshova et al, 2015 ; Shevchenko et al, 2015 ; Wu et al, 2015 ). This suggests that sulfation at the C2 of Fuc residues is not crucial for AT-binding.…”

“…Marine algae oligosaccharides, polysaccharides and their derivatives together with several animal-originated polysaccharides were prepared based on the previous studies ( Ciancia et al, 2005 ; Wang et al, 2010 ; Anastyuk et al, 2012 ; Jin et al, 2012 , Jin et al, 2013 , Jin et al, 2017 , Jin et al, 2018a , Jin et al, 2018b , Jin et al, 2019 , Jin et al, 2020a , Jin et al, 2020b , Jin et al, 2020c , Jin et al, 2020d ; Rodriguez-Jasso et al, 2013 ; Menshova et al, 2015 ; Shevchenko et al, 2015 ; Wu et al, 2015 ). They were summarized in Scheme 1 , Table 1 with proposed structures in Figure 1 and more details can be found in Supplementary Table S1 ( Supplementary data 1 ).…”

“…SJ and SJ-D were separated by anion-exchange chromatography on a DEAE-Bio Gel agarose FF (6 cm × 40 cm) eluted with water (5 L), 0.5 M NaCl (5 L) (Fraction W: SJ-W and SJ-D-W), 1 M NaCl (5 L) (Fraction I: SJ-I and SJ-D-I) and 2 M NaCl (5 L) (Fraction S: SJ-S and SJ-D-S) [ 14 ]. Autohydrolysis was carried out by methods described in previous studies [ [18] , [19] , [20] , [21] , [22] , [23] , [24] ]. Two types of autohydrolyzed polysaccharides, including higher molecular weight, H-type fractions (SJ-D-W-H, SJ-D-I-H and SJ-D-S-H) and lower molecular weight, L-type fractions (SJ-D-W-L, SJ-D-I-L and SJ-D-S-L) were obtained [ 14 ].…”
