Automated CD14+ monocyte collection with the autoMNC program of the COM.TEC cell separator

Abstract: The autoMNC program reduced the buffy coat (BC) volume and RBC yields in products compared to the standard MNC program. Increasing the CV (standard MNC program) reduced residual PLTs, RBCs, and the BC volume of MNC harvests. The donor WBC predonation count was a good predictor for the monocyte yield of products.

“…Defining the MNC apheresis settings, a central requirement was to achieve comparability of target cell yields by similar processing times of the apheresis procedures. Thereby, those settings for the autoMNC programme of the COM.TEC were applied, which have been evaluated before and assure apheresis products, which comply with the customers′ requirements for an optimum MNC harvest and subsequent cell processing . Hence, apheresis procedures with the COM.TEC were accomplished with an inlet blood flow rate of 50 ml per minute during the separation phase and 15 collection cycles with a cycle volume of 350 ml.…”

“…Regarding the RF values, the values of Punzel et al [10] for WBCs, MNCs, and lymphocytes were nearly twice as high when compared to our values. The same applies to the RF values for CD45+, CD3+ and CD14+ cells presented by Strasser et al 2005 [15] and 2007 [18]. It is likely that all these diverse observations in cell loss and the RF may be due to the different calculation formulas used.…”

“…Taking the fluid balance into account, we demonstrated a perfect negative correlation between the cell loss and recruitment for all cell populations - the higher the recruitment the lesser the cell loss of the respective cell population in the CBC. Strasser et al [18] demonstrated likewise an inverse correlation between the WBC or PLT loss and the WBC or PLT recruitment, respectively. But in contrast to our data, the perfect correlation of both variables could not be shown.…”

“…For this therapeutical approach, important quantities of monocytes are usually required to get enough DCs for vaccination purposes. The plasma needed for the cell cultures [71] is concurrently collected with the mononuclear cells [79–83].…”
