European eels (Anguilla anguilla L.) has a great dispersion from Sargasso Sea to across the Europe and North Africa. Their population have decreased dramatically over the years and listed in IUCN as critically endangered. Stock estimation is one of the important issues in order to sustainable management of this species. Last decades, researchers have focused various studies which based on monitoring of various life stages of eels. Manual counting methods with ladder traps is commonly used based on collection of glass and elver stages of eels from the natural habitats while their migration to upward of streams. During their special migration pattern, a counting device on the eel ladders can be designed so that the eels may be counted while passing through this device. The offered prototype was designed for eel ladders which was briefly, powered by an open-source electronics platform, Light Dependent Resistor (LDR) and 650 nm Laser light were used to count elver eels. Several trials were conducted to test this prototype and results were promising. This study aimed to explain features of prototype and working principle of elver counter. Designed prototype has several advantages such as usable with eel ladders, easy to customisation and affordable characteristics. However, this device needs some improvements in order to effective usages in the field.

While many cell culture systems are sensitive to the conditions in which cells are introduced into the system, we find that in situ differentiated tube-shaped microfluidic organoids have a particularly high sensitivity. Preliminary experiments using conventional seeding techniques revealed that biases in initial cell number and distribution dramatically impacted organoid shape and behavior downstream. Residual flows during seeding further complicated the process, dispersing cells to undesirable locations within the chip. To address this problem, a a robotic seeding system for controlling the process of inserting cells into microfluidic chips was developed. Environmental control of temperature, CO2, and humidity was implemented by modifying a commercial Arduino-controlled incubator. An eight-channel syringe pump controlled flow to eight cell dispensers, while a vertical leadscrew stage raised and lowered them, and a set of stackable flexure micromanipulators individually controlled the X and Y position of each cell dispenser. The flexure manipulators were 3D printed, driven by low-cost motors and electronics, and required little assembly and no alignment, resulting in a cheap and scalable method of controlling a dense array of micromanipulators. A dual objective microscope on a motorized gantry used an oblique lighting system to observe the seeding process, allowing for real-time interventions or passive observation of automated protocols. The robotic cell seeding system provided a platform for optimizing a sensitive process towards increasing the repeatability and physiological relevance of tube-shaped microfluidic organoids.

To improve particle radiotherapy, we need a better understanding of the biology of targeted and non-targeted radiation effects. This is particularly important in the field of heavy ion radiation therapy where global effects are observed, yet only a subset of cells receive a high energy deposition from ion tracks. To allow real-time 3D imaging of living biological samples during and after irradiation with a focused ion beam, we have integrated a high-speed light sheet fluorescence imaging system (swept confocally aligned planar excitation, SCAPE microscopy) into the focused ion beam irradiation platform (FIBIP) at the Columbia University Radiological Research Accelerator Facility (RARAF). The RARAF FIBIP is centered on a 7 T superconducting solenoid magnet, which focuses an ion beam, generated by RARAF’s ion accelerators, to a field size of between 5 mm and 2 µm. An integrated SCAPE volumetric imaging system with associated environmental control allows sub-second observation of 3D cell cultures sample over extended periods before, during, and after radiation treatments. The developed system enables exploration of the mechanistic effects of ion radiotherapy at the cellular and tissue levels. Here, we report details of the integrated system, and show an example of intracellular calcium signaling following ionizing radiation in U87 human glioblastoma 3D cultures using the integrated system.
