A method has been developed for determination of erythrocyte glucose consumption at a wide range of glucose concentrations. The method is based on paper chromatographic separation and quantitation of C'4-glucose and its labelled metabolites after incubation of intact cells.
Solution and chemicalsThe following buffered solutions were used (all chemicals of reagent grade): A. Phosphate buffered solution, pH 7.4 at 37" C: 0.108 M NaCl, 0.0039 M KCI, 0.005 M MgCL and 0.0202 M phosphate buffer. AB. As above but with 0.0039 M NaCl and 0.108 M KCl. B. Tris buffered solution, pH 7.4 at 37" C: 0.108 M NaCl, 0.0039 M KCl, 0.005 M MgCl., 0.001 M phosphate buffer and 0.0288 M Tris-C1 buffer. C. Phosphate buffered solutions with different pH values.
