Autometallography and metallothionein immunohistochemistry in hepatocytes of turbot (Scophthalmus maximusL.) after exposure to cadmium and depuration treatment

Abstract: In this study, autometallography and immunohistochemistry were used to localize and quantify cadmium and metallothionein (MT) levels, respectively, in cellular compartments of turbot liver on exposure to cadmium for 7 days and further depuration treatment for 14 days. Metals weakly bound to proteins (i.e. MTs) in hepatocyte lysosomes were visualized as black silver deposits (BSDs) using a light microscope. With the aid of a newly developed immunohistochemical procedure, MTs were localized and semi-quantified i… Show more

“…Positive immunohistochemical staining for MTs has been reported in the liver and trunk kidney of a few species of fish, e.g., the carp and turbot (Kito et al, 1986;Amaral et al, 2002). The immunoreactivity findings in the mandarin fish are similar to those in the carp and turbot, but the extent and intensity of the immunohistochemical staining are lower in the mandarin fish.…”

“…Albumin is the most abundant plasma protein; it suppresses the adsorption of other proteins that may empower aggravation and bacterial colonization [27]. e molecular weight of BSA is approximately 6-7 kDa [28]. In this study, the amount of fibronectin adsorbed was greater than the amount of albumin adsorbed, suggesting that adsorption quantity was related to the molecular weight of the injection solutions.…”

“…All four metals have been shown to induce MT expression, either directly or indirectly by altering the concentrations of Cu and/or Zn in the cytosol of cells from different organs (Bracken and Klaassen 1987;Jeffery et al 1987;Bobillier-Chaumnont et al 2006;Huang et al 2007). The synthesis of MT is efficiently induced in several organs to cope with the excessive exposure to metals, acting as a way of neutralizing and eliminating those elements (Kägi and Kojima 1987;Mullins and Fuentealba 1998;Amaral et al 2002;Bobillier-Chaumont et al 2006). Without MT synthesis, damages in the affected organ produced by the excess of metals could be larger than those occurring in the presence of MT, since detoxification would be delayed (Jia et al 2004).…”

“…After establishing the criteria to state the consensus basis, a previous trial was done with no significant differences among the results obtained by the four observers. According to a method previously described (Tuccari et al 2000;Amaral et al 2002), the percentage of stained cells (staining score) was graded as follows: 0 (no staining); 1 (>0 to 5%); 2 (>5 to 50%); 3 (>50%). Additionally, an intensity-distribution index (IDI) was calculated by multiplying, for each case, the staining score by the staining intensity (weak = 1; moderate = 2; strong = 3).…”
