Autophagy in Protozoan Parasites: Trypanosoma Brucei as a Model

Abstract: " To gain further and more complete understanding of the mechanisms and functions of autophagy, both conserved and evolutionarily divergent, study of more divergent model systems is needed. " Autophagy is an intracellular pathway that clears macromolecules, organelles and intracellular pathogens through autophagosomes and lysosomes upon nutrient limitation, infections and other physiological/pathological conditions. Autophagy has been best studied in budding yeast and mammals, revealing highly conserved machin… Show more

“…For example, the acidification of acidocalcisomes (organelles involved in polyphosphate metabolism) was directly related to autophagic regulation. In T. brucei , autophagy was blocked by the impairment of acidocalcisome biogenesis, suggesting that this organelle participates in this pathway 126,127 .…”

“…Conversely, among the amino acids are not only those that protect against autophagy, but also ones that, by the trypanosome’s starvation for them, induce the process, similarly to what has been reported for other eukaryotes. For example, He and collaborators described that amino acid starvation can trigger autophagy independently of the AMP-activated-Protein-Kinase-mediated energy sensing mechanisms [ 317 , 318 ]. Further studies are required to unravel the details of the pathways involved.…”

“…Form I and II of ATG8 can be separated by SDS-PAGE in the presence of urea, and form II (strong hydrophobicity) migrates more rapidly than the form I. Therefore, the form II level can be evaluated by calculating the ratio of form II/form I, and the number of autophagosomes is estimated 5 .…”
