Axon diodes for the reconstruction of oriented neuronal networks in microfluidic chambers

Abstract: Various experimental models are used to study brain development and degeneration. They range from whole animal models, which preserve anatomical structures but strongly limit investigations at the cellular level, to dissociated cell culture systems that allow detailed observation of cell phenotypes but lack the highly ordered physiological neuron connection architecture. We describe here a platform comprising independent cell culture chambers separated by an array of "axonal diodes". This array involves asymme… Show more

“…11,12 These systems have been used to great effect to study axon degeneration and regeneration following chemical 12,13 or laser [13][14][15] axotomy, tauopathy 16 and viral 17,18 dissemination, and mRNA localization in axons. 11 The systems have also been adapted for engineering the polarity of the synaptic junction using geometric diodes 19 or for synaptogenesis screening using HEK293 cells disguised as post-synaptic structures by recombinant decoration with neurolignin-1. 20 Despite the many benefits of compartmentalized neuron culture platforms there is still great scope for improvement.…”

“…We have previously designed and developed a μFD system to polarize neuronal networks using funnel-shaped micro-channels, also called axonal diodes, allowing the reconstruction of an orientated functional cortico-striatal neuronal network in vitro [8]. In the present study, we used a similar μFD-based approach to study the molecular mechanism of neurodegenerative processes in compartmentalized neurons and neuronal networks.…”

“…Microfabricated devices have been developed to facilitate both applied and basic research concerning the biology of cells and tissues. [25][26][27][28][29] The successful reconstitution of organ-level lung tissue on a microfluidic chip indicates that biomimic microsystems could potentially serve as replacements for animal testing. 28 Reconstruction of a blood vessel model has received particular attention in microfluidic technology as variable-sized channels are easily created to mimic vessel cavities.…”
