Baculovirus Expression of Chimeric Hepatitis B Virus Core Particles with Hepatitis E Virus Epitopes and Their Use in a Hepatitis E Immunoassay

Abstract: Two hepatitis B core proteins bearing the immunodominant region of the hepatitis E virus (HEV) capsid protein, one at the C terminus of hepatitis B virus core antigen (HBcAg) and the other within the HBcAg immunodominant loop, were constructed. Both chimeric proteins exhibited HEV reactivity, but only the first construct retained HBcAg reactivity. The second construct was used to develop an anti-HEV test which is equivalent to a commercial test for the detection of anti-HEV immunoglobulin G (IgG) but is more s… Show more

“…VLPs closely fit an ideal immunogen. One promising application for the production of non-pathogenic VLPs is to act as most effective immunogens to induce protective immunity and to develop immunogen-based assays [23][24][25]. Another possible application is to define the possible packaging signal of SARS CoV.…”

“…Complexes obtained by direct interaction (see below) were analyzed by electrophoresis on 1% agarose gel followed by ethidium bromide staining. Hepatitis B core particles obtained by expression in insect cells of the Core gene deleted of its 39 C-terminal amino acids (HBcv) was used as control [23].…”

“…Thus HBcAg has opened the path to the development of more potent therapeutic and preventive vaccines against HBV infection. In addition, HBcAg has become a favourite choice as immunogen carrier for various foreign epitopes [7,8] owing to its ability to self‐assemble in homologous and heterologous expression systems [9].…”
