Beneficial and Pathogenic Arabidopsis Root-Interacting Fungi Differently Affect Auxin Levels and Responsive Genes During Early Infection

Abstract: Auxin (indole-3-acetic acid, IAA) is an important phytohormone involved in root growth and development. Root-interacting beneficial and pathogenic fungi utilize auxin and its target genes to manipulate the performance of their hosts for their own needs. In order to follow and visualize auxin effects in fungi-colonized Arabidopsis roots, we used the dual auxin reporter construct DR5::EGFP-DR5v2::tdTomato and fluorescence microscopy as well as LC-MS-based phytohormone analyses. We demonstrate that the beneficial… Show more

“…Similarly, in Arabidopsis, inoculation with Trichoderma virens increases the transcript accumulation of auxin genes [54]. It was also observed that P. indica significantly stimulates the expression of auxin-responsive reporter genes both in lateral root primordia and the root elongation zone of Arabidopsis within one day of inoculation [55].…”

“…Auxin and cytokinin are major growth hormones involved in plant growth promotion response of P. indica in diverse plants (Sirrenberg et al, 2007; Vadassery et al, 2008; Meents et al, 2019). P. indica needs also induces the GA biosynthesis during colonization (Cosme et al, 2016).…”

“…Auxin and cytokinin are major growth hormones involved in plant growth promotion response of P. indica in diverse plants (Xu et al ., 2018). P. indica induces a rapid increase in auxin levels during early recognition phases which is crucial for reprogramming root development (Meents et al ., 2019). Auxin level increased upon P. indica infestation in Arabidopsis roots (Vadassery et al .,2008) and P. indica also produces IAA in liquid culture (Sirrenberg et al ., 2007).…”

“…Moreover, genes in the response to auxin stimulus were observed in both plants, and auxin mediated signaling pathway genes have also been differentially expressed in pepper ( Table 4 ). Root-interacting beneficial fungi Piriformospora indica and Mortierella hyaline induced auxin-responsive genes in the roots of their host [ 44 ]. An intricate crosstalk between these hormones seems to take place in the responses triggered in both pepper and tomato.…”
