Bimetallic Assembled Silver Nanoparticles Impregnated in Aspergillus fumigatus Extract Damage the Bacterial Membrane Surface and Release Cellular Contents

Abstract: The bactericidal effects of nanomaterials play an essential role in cytoplasmic leakage, leading to bacterial cell death. In this study, silver nanoparticles (AgNPs) were synthesized using a fungal extract of Aspergillus fumigatus (A. fumigatus). The physicochemical properties of the bare and myco-synthesized AgNPs (MS-AgNPs) were examined by X-ray diffraction (XRD), Fourier transform infrared (FT-IR) spectroscopy, ultraviolet–visible (UV–vis) spectroscopy, scanning electron microscopy (SEM), energy dispersive… Show more

“…The volatile oils were serially diluted in 5% (DMSO) Dimethyl sulfoxide solution containing 0.1% Tween 80 ( v / v ) (10 mg/mL), and then, 50 μL of each dilution was added to wells in a microtiter plate containing 100 μL TSB. Fifty microliters of adjusted microbial inoculum (10 6 cells/mL) were added to each well, and then the microtiter plates were incubated in the dark at 37 °C for 24 h. After incubation, 100 μL of freshly prepared XTT were added and incubated again for 1 h at 37 °C [ 36 ]. Colorimetric variation in the XTT assay was measured using a microtiter plate reader at 492 nm.…”

“…The agar disc diffusion method [ 31 ] facilitated the determination of antibacterial activity in all of the compounds [ 32 , 33 ]. Four bacterial strains including two Gram-negative ( Staphylococcus aureus, Escherichia coli ) and two Gram-positive ( Bacillus pumilis, Salmonella typhi ) were tested.…”
