Bimetallic metal–organic frameworks for tumor inhibition via combined photothermal-immunotherapy

Abstract: Herein, we report the design of therapeutic nanoparticles by encapsulating photosensitizers and aluminum ions into metal-organic frameworks. The nanoparticles could significantly inhibit the growth of primary and rechallenged tumors by...

“…These strategies include ICD, immune agonists and cancer vaccines, immune checkpoint blockade and tumorspecific mAbs, and small molecule immunosuppressants. 207 Li et al 208 investigated a novel NP. Aluminum ions and photosensitizers (i.e., indocyanine green, ICG) were encapsulated into zeolitic imidazolate framework 8 (ZIF-8) NPs (ICG@ ZIF8(Al) NPs) for PTT in combination with immunotherapy to eradicate cold tumors.…”

“…The synthesized ZIF‐8(Al) can be used to encapsulate ICG, and the aluminum ion itself can be used as an immune adjuvant to amplify the tumor‐specific immune response. “…Porous metal–organic frameworks (MOFs) composed of organic ligands and metal ions have been widely used for catalysis and biomedical applications. Therein, zeolitic imidazolate framework-8 (ZIF-8) is one of the mostly studied MOFs due to the ease of synthesis procedures, pH-responsiveness, and their versatility for the encapsulation, protection and delivery of biomacromolecules. In our previous work, we reported the preparation of ZIF-8 NPs by using poly(ethylene glycol) (PEG) as the mineralizer, which enabled the co-encapsulation of either positively or negatively charged biopolymers and significantly improved the colloidal dispersity of ZIF-8 NPs. The polymer-loaded ZIF-8 NPs are promising templates for the assembly of polymer NPs.…”
