Treatment of this lethal infection is lengthy, and in some 45 patients therapy is not curative and patients require lifelong therapy. Fundamental 46research in this yeast is needed so that we can understand mechanisms of infection 47 and disease and ultimately devise better therapies. In this work we investigated a fungal 48representative of the annexin family of proteins, specifically in the context of virulence 49 and mating. We find that the cryptococcal annexin does not seem to be involved in 50 virulence or mating but affects generation of titan cells, enlarged yeast cells that are 51 detected in the lungs of mammalian hosts. Our data provides new knowledge in an 52 unexplored area of fungal biology. 53 annexins are phylogenetically closer to Dictyostelium discoideum amoeba model 131 organism than to human or plant annexins, and consequently, fungal and amoeba 132 annexins are grouped in clade C. The sequence of fungal annexins differs significantly 133
