Biolytic extraction of poly(3-hydroxybutyrate) from Bacillus megaterium Ti3 using the lytic enzyme of Streptomyces albus Tia1

Abstract: The applicability of Streptomyces sp. cell lytic enzymes for devising a simple and competent biological polyhydroxyalkanoate (PHA) recovery approach from Bacillus megaterium cells was investigated. B. megaterium strain Ti3 produced 50% (w/w) PHA using glucose as carbon source. The intracellular PHA was recovered employing a non-PHA accumulating actinomycetes (Tia1) identified as Streptomyces albus, having potent lytic activity against living and heat inactivated B. megaterium. Interestingly, maximum biomass (2… Show more

“…The enzymatic technique typically includes a thermal pre-treatment of the biological material, followed by enzymatic hydrolysis, which is then treated with surfactants before being decolored with H 2 O 2 [102]. A proteolytic and caseinolytic enzyme produced from Streptomyces albus has been utilized to extract PHA from B. megaterium with a high purity of 90-93% [105].…”

“…[ 164 ] Lytic enzymes of S. albus Tia1 (2.5 g L −1 ), when used with PHA producing B. megaterium Ti3, resulted in polymer recovery up to 0.55 g g −1 CDW. [ 165 ] Lysozyme‐inducing system constructed in recombinant P. putida JBOi could induce cell lysis improving PHA recovery up to 75%. [ 166 ] Autolysis of cell membrane was induced in recombinant P. putida BXHL with endolysin ( Ejl ) and holin ( Ejh ) genes of pneumococcal bacteriophage Ej‐1 which recovered 0.86 ± 0.09 g L −1 PHA.…”

“…Israni et al, harnessed the high lytic activity of Streptomyces albus Tia1 for isolation/purification of PHA from PHA-producing cells [ 313 ]. In one approach, S. albus and PHA-producing cells were co-inoculated.…”

“…Conditions resulting in the maximum activity for cell lysis of PHA were the addition of a concentrated S. albus culture filtrate (33.3 mL) from a 100 mL cultivation to B. megaterium (220 mg) with incubation at 40 °C and pH 6. By co-inoculation of S. albus and PHA producing B. megaterium , the PHA yield reached 0.55 g/g which was similar to treatment with sodium hypochlorite for PHA recovery [ 313 ]. In contrast, comparison of the enzyme based with co-inoculation showed that the former resulted in a 1.74-fold increase in PHA yield [ 313 ].…”
