Objectives Zein nanoparticles (Zein NPs) were used as a hydroxyapatite (HA) biomineralization template to generate HA/Zein NPs. Doxorubicin hydrochloride (DOX) was loaded on HA/Zein NPs (HA/Zein‐DOX NPs) to improve its pH‐sensitive release, bioavailability and decrease cardiotoxicity. Methods HA/Zein‐DOX NPs were prepared by phase separation and biomimetic mineralization method. Particle size, polydispersity index (PDI), Zeta potential, transmission electron microscope, X‐ray diffraction and Fourier‐transform infrared spectroscopy of HA/Zein‐DOX NPs were characterized. The nanoparticles were then evaluated in vitro and in vivo. Key findings The small PDI and high Zeta potential demonstrated that HA/Zein‐DOX NPs were a stable and homogeneous dispersed system and that HA was mineralized on Zein‐DOX NPs. HA/Zein‐DOX NPs showed pH‐sensitive release. Compared with free DOX, HA/Zein‐DOX NPs increased cellular uptake which caused 7 times higher in‐vitro cytotoxicity in 4T1 cells. Pharmacokinetic experiments indicated the t1/2β and AUC0–t of HA/Zein‐DOX NPs were 2.73‐ and 3.12‐fold higher than those of DOX solution, respectively. Tissue distribution exhibited HA/Zein‐DOX NPs reduced heart toxicity with lower heart targeting efficiency (18.58%) than that of DOX solution (37.62%). Conclusion In this study, HA/Zein‐DOX NPs represented an antitumour drug delivery system for DOX in clinical tumour therapy with improved bioavailability and decreased cardiotoxicity.

Microencapsulation is considered an efficient technique to protect functional materials from oxidization while enabling controlled release. In this study, anti‐solvent precipitation was used to prepare zein‐based microcapsules loaded with Artemisia argyis essence (AAE@ZMs). The impact of different drying methods, namely vacuum drying, freeze drying, and spray drying on AAE@ZMs was evaluated. Quality of AAE@ZMs was evaluated by the determination of color, moisture content, bulk density, chemical structure, and morphology evaluation. Freeze dried AAE@ZMs (F‐AAE@ZMs) and vacuum dried microcapsules (V‐AAE@ZMs) respectively showed stronger water and oil absorption capacities. The residual content of Artemisia argyis essence (AAE) in V‐AAE@ZMs was higher than those in F‐AAE@ZMs and spray dried microcapsules (S‐AAE@ZMs) after continuously releasing for 120 h. Meanwhile, heated from 30 to 600°C, the residues of V‐AAE@ZMs and F‐AAE@ZMs were lower than those of S‐AAE@ZMs. Therefore, drying methods greatly affected key quality parameters of AAE@ZMs. This study provides guidance on the use of drying methods in microcapsules delivery systems with zein or other materials.

Biopolymers are playing a vital role in biomedical applications. Among them, protein‐based biopolymers are utilized for the fabrication of tissue‐engineering constructs, therapeutic molecule delivery carriers, emulsifiers, and food packaging materials. Wide ranges of proteins are extracted from animal or plant sources and are being utilized for the fabrication of scaffolds for regenerative tissue‐engineering application. Here, an overview about the protein structure, extraction procedure, solubility, and various formulation‐based proteins found in the literature are discussed. Biopolymers display several advantages such as biocompatibility and degradability by enzymes. Methods to overcome the disadvantages of these proteins such as immunogenicity, antigenicity, and solubility are reported. Various crosslinking reagents specific to protein chemistry are discussed as well.
