Bronchial Epithelial Cells Accumulate Citrate Intracellularly in Response to Pneumococcal Hydrogen Peroxide

Abstract: Community-acquired pneumonia is an infection of the lower respiratory tract caused by various viral and bacterial pathogens, including influenza A virus (IAV), Streptococcus pneumoniae, and Staphylococcus aureus. To understand the disease pathology, it is important to delineate host metabolic responses to an infection. In this study, metabolome profiling of mono-and coinfected human bronchial epithelial cells was performed. We show that IAV and S. aureus silently survive within the cells with almost negligible… Show more

“…All infections were performed in RPMI1640 complete media. 1 × 10 5 monocyte-derived DCs (moDCs) were infected with H1N1 at a multiplicity of infection 0.1 for 24 h [ 30 ]. To warrant comparable initial bacterial infection rates, moDCs were infected at multiplicity of infection 10 for 3 h with pneumococci and 1 h with staphylococci.…”

“…Infections of monocytes and macrophages were performed in RPMI1640 media supplemented with 10% FCS. Furthermore, 1 × 10 5 monocytes or 2 × 10 5 monocyte-derived macrophages were infected with H1N1 at an MOI of 0.1 for 24 hours 41 . The MOI was chosen based on previous results, showing equal viral loads for a range of tested MOIs 18,41 .…”

“…41 The MOI was chosen based on previous results, showing equal viral loads for a range of tested MOIs. 18,41 Macrophages and monocytes were infected with SA113 at MOIs of 10 and 1, respectively. After one hour, the medium was removed, and extracellular bacteria were killed by addition of RPMI1640 containing 400μg/mL gentamicin or 2-μg/mL lysostaphin (both Sigma-Aldrich).…”

“…Stable isotopic labeled internal standard compounds were used as described before. [32] Absolute concentrations of metabolites were determined with a calibration curve ranging from 0.05 to 100 nmol/ sample for each metabolite. All metabolite concentrations were normalized to the wet weight of colonies.…”
