Bupivacaine hydrochloride complexation with some α- and β-cyclodextrins studied by potentiometry with membrane electrodes

Abstract: Membrane electrodes selective to bupivacaine cations were developed and those with PVC-dibutylphthalate membrane containing sparingly soluble bupivacaine phosphotungstate appeared to be the most suitable. Inclusion complexation of bupivacaine cations with cyclodextrins was studied by potentiometric measurements of the free bupivacaine cation concentration in aqueous solutions of bupivacaine hydrochloride with cyclodextrin using the prepared electrodes. Native alpha-cyclodextrin (alpha-CD) and beta-cyclodextrin… Show more

“…21 The corresponding values for a-CD and b-CD are 102 ± 10 and 63 ± 5 dm 3 mol -1 , respectively. 22 With b-CD, the binding constant decreases for the diprotonated form of procaine (1.4 dm 3 mol -1 ), while it increases for the neutral forms of procaine (from 3.0 ¥ 10 2 to 1.5 ¥ 10 3 dm 3 mol -1 ) 9h and tetracaine (from 1.36 ¥ 10 3 to 6.60 ¥ 10 3 dm 3 mol -1 ). 9j With a-CD, only the neutral form of procaine binds, with a stability constant of 120 dm 3 mol -1 at pH 10.4.…”

“…Furthermore, we assumed that two water molecules are situated inside the cavity of each R-CD, and two water tetramers surrounding it below or above the cavity, somehow similar to the R-CD down (larger cavity) or up (smaller cavity) preferred structures for the hexahydrate species found in our previous work. 26 We can also rewrite the right side of eq 2 as a complex of R-CD dimer, a H 2 O dimer inside the cavity of each CD and two water tetramers outside, i.e., 4 . So, nine distinct spatial arrangements were obtained.…”
