Ca2+-Regulated Photoproteins: Effective Immunoassay Reporters

Abstract: Ca2+-regulated photoproteins of luminous marine coelenterates are of interest and a challenge for researchers as a unique bioluminescent system and as a promising analytical instrument for both in vivo and in vitro applications. The proteins are comprehensively studied as to biochemical properties, tertiary structures, bioluminescence mechanism, etc. This knowledge, along with available recombinant proteins serves the basis for development of unique bioluminescent detection systems that are “self-contained”, t… Show more

“…We examined binding affinities of individual aptamers by the solid-phase bioluminescent sandwich microassay. A bioluminescent photoprotein obelin was chosen as a fast, triggerable, and highly sensitive reporter for the assay [20]. 3'-Biotinylated aptamers were immobilized on the streptavidinactivated surface (thus, employed as capture components of the sandwich) and titrated with total or glycated Hb.…”

“…This is a basis for medico-biological application of the photoprotein bioluminescence in different types of diagnostics: for monitoring the Ca 2 + content, protein location in cells and tissues, and others. Additionally, bioluminescence of photoproteins is used as an intracellular marker [15,16]. These are reasons for intensive investigations of the photoprotein bioluminescence mechanism.…”

“…Aequorin as a Ca 2+ -activation type is used for its unique advantages, such as detection within few seconds at the attomole level, high sensitivity and low background noise, high quantum yield, harmless application, lack of cellular toxicity, being active at physiological pH, hazard-free handling and noninvasive nature [16][17][18].…”
