Calcitonin-Derived Carrier Peptides

Abstract: The recent discovery of carrier peptides offers new opportunities to translocate several bioactive molecules into the cytoplasm. Previous studies have shown that human calcitonin (hCT) and selected C-terminal sequences translocate in nasal epithelium. Moreover, the hCT(9-32) fragment was found to internalize efficiently a number of substances like fluorophores, nucleic acids or the enhanced green fluorescent protein (EGFP). In order to understand the uptake mechanism interactions of hCT(9-32) with membrane mod… Show more

“…Owing to a high permeability through the nasal epithelium, it is applied as an intranasal therapeutic . Structure−activity studies revealed that the truncated sequence hCT(9−32) can translocate through cell membranes, although the calcitonin receptor activating N-terminus (aa 1−8) of hCT is lacking , . By using hCT(9−32), efficient transport of different cargoes into cells like the antiproliferative drug daunorubicin, fluorophors, or even large proteins has been achieved , .…”

“…9 Recently, truncated sequences of the human peptide hormone calcitonin (hCT) were successfully applied as delivery vehicle for different substances, like proteins, oligonucleotides, and small organic molecules. 10 Since then, very promising branched structures have been developed and have served for the delivery of plasmid DNA as well as nanoparticles. 11,12 The side sequence of these branched peptides contains positive charged amino acids for complexation of negatively charged DNA or RNA molecules.…”

“…Recently, CPPs based on the human calcitonin have been described . Calcitonin is a 32 amino acid long peptide hormone that is physiologically secreted by the thyroid gland and involved in the regulation of the calcium homeostasis.…”
