Capillary electrochromatography and nano‐liquid chromatography coupled to nano‐electrospray ionization interface for the separation and identification of estrogenic compounds

Abstract: Nano-LC and CEC were coupled to MS through a nanospray or a pressurized liquid-junction interface for the simultaneous separation and determination of 11 estrogenic compounds. Different stationary phases, that is, phenyl, C18, and C18 bidentate silica hydrate, were studied. For both techniques, the phenyl stationary phase was the best option, considering separation efficiency, selectivity, and resolution. Under the optimized conditions, the baseline separation of the target compounds (including estradiol and z… Show more

“…D'Orazio et al. proposed the hyphenation with MS by nanospray or pressurized liquid‐junction (L‐J) interface for the separation of 11 estrogenic compounds. Under the optimized conditions, the baseline separation of the target compounds was achieved in less than 20 min in nano‐LC‐MS and less than 13 min in CEC‐MS.…”

“…The cost and the still low availability of commercial capillary columns force most researchers to prepare packed columns by themselves (Fanali et al, 2012a,b;Blue and Jorgenson, 2015;D'Orazio et al, 2016b). However, packing and retaining an SP in narrow-bore capillaries can be quite difficult and requires dedicated apparatuses, as well as a certain experience, to warrant a good stability and reproducibility between columns.…”

“…The University of San Pablo published 3 papers in 2016 [125][126][127] using multiple mass spectrometry based untargeted technologies [LC-MS, CE-MS and GC-MS] to create large data rich metabolomics profiles in plasma [125], serum [126] and from cell culture [127] to identify biomarkers in obesity and hypercholes- terolemia. D'Orazio et al [121] compared nano LC and CE-MS to quantitatively measure estrogenic metabolites from mineral water. These nano techniques were used to minimize sample volumes and reagent costs.…”
