Capparis Spinosa L. promotes anti-inflammatory response in vitro through the control of cytokine gene expression in human peripheral blood mononuclear cells

Abstract: BackgroundCapparis Spinosa L. is an aromatic plant growing wild in dry regions around the Mediterranean basin. Capparis Spinosa was shown to possess several properties such as antioxidant, antifungal, and anti-hepatotoxic actions. In this work, we aimed to evaluate immunomodulatory properties of Capparis Spinosa leaf extracts in vitro on human peripheral blood mononuclear cells (PBMCs) from healthy individuals.ResultsUsing MTT assay, we identified a range of Capparis Spinosa doses, which were not toxic. “…Peripheral blood mononuclear cells “PBMCs” were separated from fresh venous blood collected from healthy donors as previously described [ 19 ]. PBMCs isolation was performed by centrifugation on Ficoll- histopaque 1077 (Sigma, USA), at 900 g for 25 min at 18–20 °C, then washed three times by centrifugation, and suspended in RPMI 1640 media (sigma, USA) with L-glutamine supplemented with 10 % heat inactivated newborn calf serum (sigma, USA), 26.3 g/l penicillin and 4.2 g/l streptomycin.…”

“…Cytotoxicity of A.S. extract to PBMCs was evaluated by MTT (3-(4, 5 dimethylthiazol-2-yl)-2, 5 diphenyltetrazolium bromide) assay [ 19 , 20 ]. The test was performed in 96-well plates (Hiwaka, Japan).…”

“…Previous studies have indicated the presence of active compounds in C.spinosa such as alkaloids, lipids, flavonoids and clocosinols [5]. They have a number of biological agents such as antifungal, antifungal and liver toxicity [15].…”

“…Long-term use of opioids leads to dependence, euphoria, and tolerance. Long-term treatment or high levels of corticosteroids may result in indigestion, muscle weakness, fluid retention, stomach bleeding, osteoporosis, obesity, heart failure, respiratory problems, anemia, liver damage, and Stevens-Johnson syndrome (5)(6)(7). Therefore, researchers have suggested use of traditional medicine as a tried and useful method of pain and inflammation management (8).…”
