Capsular and extracellular polysaccharides of the diazotrophic rhizobacterium Herbaspirillum seropedicae Z78

Abstract: The diazotrophic endophyte Herbaspirillum seropedicae Z78 was shown to possess a capsule con taining two high molecular weight glycolipids, one of which was of a lipopolysaccharide nature. These gly colipids differed considerably in the fatty acid composition of their lipid components. The polysaccharide moiety of these glycans was composed of glucose, galactose, glucosamine, galactosamine, and a noncarbo hydrate component, butanetetraol. In the culture liquid of H. seropedicae Z78, an extracellular polysaccha… Show more

“…The analyses of H. seropedicae eps genes that code for glycosyltransferases and sugar modifying proteins (such as epimerases and phosphatases in Table S1 ) suggest that the EPS is composed of these same monosaccharides. Indeed, monosaccharide composition analysis of H. seropedicae Z67 T EPS showed galactose, glucose and mannose as constituents at a proportion of 4∶3:1, with possible substitutions with tetracarboxylic acids [53] .…”

“…Despite the presence of KDO and~40% of hydroxylated fatty acids, the EPS is a polysaccharide-lipid complex. This assumption is based on the fact that the ultracentrifugation of the EPS did not yield a transparent gel-like sediment (LPS), contrary to what was observed by Smol'kina et al (2012).…”

“…Herbaspirillum seropedicae Z78 (IBPPM 217) was from the IBPPM RAS Collection of Rhizosphere Microorganisms (http://collection.ibppm.ru). Cells were grown in a vitaminsupplemented liquid synthetic nutrient medium (Smol'kina et al 2012) at 30 ± 1°C for 24 h (until the end of the exponential growth phase).…”

“…The cells were then degreased with petroleum ether, dried with acetone, and finely dispersed. CPS-I, CPS-II, EPS-I and EPS-II were isolated as described by Smol'kina et al (Smol'kina et al 2012). All carbohydrate-containing fractions that did not give absorption between 240 and 260 nm were pooled, concentrated, and lyophilized.…”
