Carbon monoxide and bilirubin from heme oxygenase-1 suppresses reactive oxygen species generation and plasminogen activator inhibitor-1 induction

Abstract: Heme oxygenase-1 (HO-1) responds to a variety of oxidative stresses. We examined whether HO-1 expression influences pro-thrombotic processes, in which the involvement of oxidative stress has been reported. Since HO-1 knockout mice with a C57/BL6J background were not viable, embryonic cells from HO-1 deficient mice (E11.5) were used. Cell viability, the level of plasminogen activator inhibitor-1 (PAI-1) expression and reactive oxygen species (ROS) generation of HO-1 deficient cells in response to the exposures … Show more

“…35 HO-1, which is a rate-limiting enzyme that metabolizes heme accumulates in tissues because of blood red cell turnover, is correlated with the production of ROS. 36 Against this background, our purpose was to find out whether RvD1 can protect TJ proteins from disruption by Regulation of HO-1 activity as an adaptive response to oxidative stress is to ameliorate the effects of many oxidative stressors. 37 When pretreated with RvD1, mice that inhaled LPS had a high transcription and protein expression of HO-1.…”

“…Both biliverdin and bilirubin are potent antioxidants, and the inhibition of prothrombotic molecular expression by bilirubin via decreasing intracellular ROS level has been observed (39). CO is a secondary messenger with multiple biological activities.…”

“…For DCF fluorescence measurement, cells were washed with phenol red-free DMEM and pretreated with DCF diacetate (20 mmol/l) for 45 min. 17 After removing the probe, cells were incubated with H 2 O 2 (200 mmol/l) in the presence or absence of kallistatin (0.5 mmol/l) or L-NAME (200 mmol/l). DCF fluorescence was recorded (485 nm excitation, 538 nm emission) for a period of 60 min.…”

“…18,27 TGF-b can also prevent the degradation of collagen via increased PAI-1 expression. 17,28 It has been reported that ROS modulate fibroblast proliferation and their transition into matrix-producing myofibroblasts in the heart. 5,29 Furthermore, ROS induce expression of TGF-b1 and type I collagen, and lead to activation of TGF-b from its latent form.…”
