Carbonate hydroxyapatite functionalization: a comparative study towards (bio)molecules fixation

Abstract: Different methods for the functionalization of carbonate hydroxyapatite granules with free amine groups by reaction with (3-aminopropyl)triethoxysilane (APTES) have been compared in order to improve the potential for tethering of bioactive molecules to bioceramics. The combined use of tetraethoxyorthosilicate and APTES with acid catalysis resulted in an evident increase in amine surface grafting.

“…Traditional pathogen enrichment methods are based on density gradient centrifugation and cell culture, which are still used in clinical analysis (Dorn et al, 1976;Hanff et al, 1984;Lagier et al, 2015). Several promising methods have been developed to capture pathogens in a small sample volume, such as antibody-, lectin-and antimicrobial peptide-based biological methods (Bicart-See et al, 2016;Chairatana and Nolan, 2014;Pal et al, 2017;Russo et al, 2014). A small sample amount (a few hundred microliters) is used in conventional assays bcause of their intrinsic limitations.…”

“…For instance, conjugation of matricellular cues within the structure of three-dimensional scaffolds requires the definition and optimization of biofunctionalization protocols. Russo et al [5] report on a flexible and effective method for the fixation of biomolecules on scaffolds of carbonate hydroxyapatite. Following this procedure, it is virtually possible to encode any relevant biomolecular signals within materials widely used for bone tissue engineering.…”

“…Small organic molecules with ionisable functional groups such as silanols, carboxylic acids, amines, thiols or nucleotide strands display high affinity for the CaP surface [221][222][223], and can therefore be employed as ligands. When the ligands are attached to CaP NPs, their exposed functional groups can be further conjugated with the active targeting molecules.…”
