Cardiac expression of polysialylated NCAM in the chicken embryo: Correlation with the ventricular conduction system

Abstract: The neural cell adhesion molecule (NCAM) and its polysialic acid moeity (PSA) affect cellular interactions during the development of the nervous system and skeletal muscle. NCAM has also been identified in the embryonic heart of various species including humans. However, knowledge regarding the role of NCAM and its function-modulating PSA in cardiogenesis is limited. The distribution of NCAM and its PSA in the ventricular myocardium of chicken embryos was determined by indirect immunofluorescence staining. The… Show more

“…Specificity of staining was verified by the absence of stain ing in tissue sections in which PSA was removed by incu bation with endo-N or when the antibody was preincu bated with chicken PSA-NCAM. Moreover, immunoblot analyses of extract from the SCN region (and other brain areas) revealed an immunoreactive band migrating in the 150-to 300-kD range, which is characteristic of PSA staining seen in different species [15,29,31,37,38]. These results are also consistent with a previous observa tion of immunostaining for PSA in the rat SCN [15].…”

“…Assuming that the innervatinn of the heart is similar to the brain in not expressing forms of NCAM recognized by anti-MSD antibodies, then the data would suggest that at least the 185-kD form of NCAM is produced within the heart. It is tempting to speculate that the ld form of NCAM may be preferentially expressed in the conduction system, given that the polysialic moieties found on NCAM, as well as the carbohydrate epitopos recognized by monoclonal antibody HNK-1 and found on NCAM and other neural adhesion molecules, are also preferentially expressed in this region of the heart (Watanabe et al, 1992;Nakagawa et al, 1993).…”

“…Cell adhesion molecules showing homophilic interactions might play an important role in this process. The neural cell adhesion molecule (N-CAM), for example, has been found in the developing epicardium as well as in the myocardium [Burroughs et al, 1991;Lackie et al, 1991;Watanabe et al, 1992]. N-CAM knockout mice, however, apparently show no anomalies in the epicardium [Cremer et al, 1997].…”
