Cardiac, renal and uterine hemodynamics changes throughout pregnancy in rats with a prolonged high fat diet from an early age

Abstract: Objective To examine whether the cardiac, renal and uterine physiological hemodynamic changes during gestation are altered in rats with an early and prolonged exposure to a high fat diet (HFD). Methods Arterial pressure and cardiac, renal, uterine and radial arteries hemodynamic changes during gestation were examined in adult SD rats exposed to normal (13%) (n = 8) or high (60%) (n = 8) fat diets from weaning. Plethysmography, high-resolution high-frequency ultrasonography and clearance of an inulin analog wer… Show more

“…Body weight and food intake were recorded weekly from 12 wk to the end of the study. Arterial pressure, stroke volume (SV), heart rate (HR), and renal blood flow (RBF) were measured at weeks 12 , 14 , 16 , and 18 of age in isoflurane anesthetized rats ( 23 ). Cardiac output (CO) values were calculated from SV and HR.…”

“…Ultrasound measurements were performed, as previously described ( 23 ), in isoflurane anesthetized rats on a heated platform to maintain rectal temperature at 37°C. SV, HR, and RBF were evaluated using a high-resolution micro-ultrasound system (Vevo 3100, VisualSonics, Toronto, Canada) and two transducers: MX250 (axial resolution: 50 µm; frequency: 25 MHz) and MX400 (axial resolution: 75 µm; frequency: 40 MHz).…”

“…Six weeks later, as indicated above [19], this scenario would be much more dramatic, and the BT group will show higher values, being significantly different from the SD diet as well. In this sense it is well known how damage inflicted by high fat diets is cumulative with time, including developing high blood pressure [23][24][25]. No other difference was detected at a physiological level, in contrast again with the results reported at 12 weeks, when the butter-fed group displayed significant higher values not only on blood pressure but also on body weight, among other hormonal and biochemical variables measured once the animal was sacrificed, like insulin, plasma triglycerides or total cholesterol [19].…”
