Catenin delta-1 (CTNND1) phosphorylation controls the mesenchymal to epithelial transition in astrocytic tumors

Abstract: Inactivating mutations of the TSC1/TSC2 complex (TSC1/2) cause tuberous sclerosis (TSC), a hereditary syndrome with neurological symptoms and benign hamartoma tumours in the brain. Since TSC effectors are largely unknown in the human brain, TSC patient cortical tubers were used to uncover hyperphosphorylation unique to TSC primary astrocytes, the cell type affected in the brain. We found abnormal hyperphosphorylation of catenin delta-1 S268, which was reversible by mTOR- specific inhibitors. In contrast, in t… Show more

“…Specifically, the Uruguayan population is admixed, with a predominant European contribution and Native American and African contributions of 10.4 and 5.6%, respectively [113, 114]. Common genetic variation affects expression variability [115], as well as the admixed genome structure [116, 117], stressing the necessity of population-specific transcriptome analyses or, alternatively, including diversity in high-throughput population analyses.…”

“…Phosphorylation of a different serine residue on this protein is involved in the control of EMT balance. 67 On the other hand, the Wnt signaling pathway is attenuated by inhibitor proteins such as secreted frizzled related protein (sFRP) and dickkopfrelated proteins, of which sFRP1 and -2 and dickkopf-related protein 3 were identified in this study. 68,69 Transcriptional Regulation.…”

“…Of these, phosphorylation of Vimentin (VIM) at S459 by PRKCD, RPS3 at T221 by PRKDC and ubiquitination of Caveolin-1 (CAV1) at K5 have been linked DNA damage repair, cytoskeleton reorganization and endosomal transport in vitro 30 , 31 . Other sites such as phosphorylation of CTNND1 at S268 by PKCE or CTNNA1 at T654 have been associated with E-cadherin, Wnt signaling and epithelial-to-mesenchymal transition 32 , 33 . These modifications have also been associated with tumor progression together with PML ubiquitination and phosphorylation at S402 and S518, and CAP1 phosphorylation by GSK3B at T307, S308 and S310 34 37 .…”
