Cbfa-1 mediates nitric oxide regulation of MMP-13 in osteoblasts

Abstract: During bone development, osteoblast differentiation requires remodeling of the extracellular matrix. Although underlying mechanisms have not been elucidated, evidence points to the participation of the nitric oxide (NO) and cyclic guanosine 3′,5′-monophosphate (cGMP) system. Here, we detected increased matrix metalloproteinase (MMP)-13 mRNA, protein and activity, as well as increased inducible NO synthase (iNOS) and NO production during the differentiation of MC3T3-E1 osteoblasts. Transcriptional activity of t… Show more

“…In converse to the finding that RUNX2 upregulates BAX expression in the SAOS-2 cell line [108], nitric oxide (NO) treatment of the MG-63 osteosarcoma cell line induces RUNX2-mediated BCL2 expression, which promotes survival of the cells during oxidative stress [111]. NO signaling through cyclic guanosine 3′,5′-monophosphate (cGMP) may also cause site-specific phosphorylation of RUNX2 by protein kinase G (PKG), leading to upregulated transcription of the matrix metalloproteinase (MMP) gene MMP13 [112]. MMP13 is one of several members of the MMP family with important roles in cartilage degradation during endochondral ossification and later bone remodeling (reviewed in [113]).…”

“…The ROS are undoubtedly one of the most effective pathogenic mechanisms in all those bacterial pathologies which lead to bone resorption 19 and several scientifical evidences show how the oxidative killing, carried out from neutrophils against pathogenic bacteria, is frequently the first stage of bone damage; especially in some dental pathologies, such as the acute apical periodontitis (AAP), the neutrophils obtained from peripheral blood of AAP subjects shown increased production of hydrogen peroxide and superoxide anion, which is instead normalized after surgical treatment 20.…”

“…Also, the somewhat varied and relatively upstream modulation of osteoblast differentiation by MMP-1 and -13 suggest early divergence in their mechanisms of induction of osteoblast differentiation. It is plausible that alternative mechanisms such as receptor-mediated activity of MMP-1 (Hayami et al, 2010), or the intricate involvement of MMP-13 but not MMP-1 in the intrinsic physiologic functions of osteoblast differentiation (Parikka et al, 2005; Pei et al, 2006; Selvamurugan et al, 2006; Varghese et al, 2005; Zaragoza et al, 2006) as well as other unknown mechanisms may explain the differences between the observed effects of MMP-1 and -13 on early osteoblast differentiation.…”
