CDH13 is Frequently Inactivated by Promoter Hypermethylation in Pediatric Acute Myeloid Leukemia

Abstract: There is growing evidence supporting a role for tumor suppressor as targets in aberrant mechanisms of DNA hypermethylation. Methylation in the promoter of tumor suppressor always plays important roles in pediatric Acute Myeloid Leukemia (AML). CDH13 gene is a tumor suppressor involved in tumorigenesis, metastasis and apoptosis in a variety of tumors. In this study, we are trying to investigate whether CDH13 was down regulated by promoter methylation in pediatric AML. MRNA transcriptional expression levels of C… Show more

“…We checked the presence of T-cadherin epitopes on the surface of human platelets and megakaryocytes using a specific antibody (R&D, BAF3264) and flow cytometry. The anti-T-cadherin antibody precisely bound with HUVEC (positive control, cells expressing T-cadherin), MEG-01 (a megakaryoblastic cell line), and platelets, but did not bind with THP-1 (negative control, cells which do not express T-cadherin according to proteinatlas.org and [ 41 ]). We demonstrated a significant difference in binding ( p < 0.001, pairwise comparison, Figure 1 a,b) between the specific antibody and the corresponding isotype control in all types of cells.…”

“…CDH13'ün tümör baskılayıcı bir gen olarak işlev görebileceği birçok kanser türünde in vitro ve in vivo çalışmalarla gösterilmiştir. Ayrıca meme ve akciğer kanserlerinde, pituiter adenomlarda, diffüz büyük B hücreli lenfomada, nazofarengeal ve kütanöz skuamöz hücreli karsinomalarda CDH13 hipermetilasyonu varlığı belirlenmiştir (4). DAPK1, Ca 2+ /kalmodulin bağımlı, sitoiskelet ilişkili bir protein kinazdır.…”
