Cell apoptosis, autophagy and necroptosis in osteosarcoma treatment

Abstract: Osteosarcoma is the most common primary bone tumor in children and adolescents. Although combined therapy including surgery and multi-agent chemotherapy have resulted in great improvements in the overall survival of patients, chemoresistance remains an obstacle for the treatment of osteosarcoma. Molecular targets or effective agents that are actively involved in cell death including apoptosis, autophagy and necroptosis have been studied. We summarized how these agents (novel compounds, miRNAs, or proteins) reg… Show more

“…Myosins are molecular motor proteins that bind to actin to perform functions such as motility, contractility, and invasion and metastasis in cancer . Myosin 1b is a member of class I myosin that promotes invasion in cervical cancer, and lymph node metastasis of head neck squamous cell carcinoma .…”

“…Apoptosis, necroptosis, and autophagy are classified as programmed cell death, an integral process to maintain a homeostatic circumstance in organisms [22]. The extrinsic and intrinsic pathways are two well-studied mechanisms that contribute to the execution of apoptosis (Figure 1; [10,11,12,13,14,15]).…”

“…A combinational application of neoadjuvant chemotherapy and consequent surgical resection significantly increased the five-year survival rate of patients without metastasis to approximately 60%-70% [3]. However, the outcome is disappointing in metastatic patients, with a five-year survival rate of only 5%-20% [4, 5]. Additionally, severe side effects and drug resistance are common.…”
