Cell features in pancreas of prediabetic and diabetic rabbits after wirsung duct ligation. Histochemical and histoenzymatic studies

Abstract: Kinetic studies of the histochemical and histoenzymatic behavior of rabbit pancreatic parenchymas were performed 5, 30 and 90 days after Wirsung duct ligation. In control pancreas, some enzyme activities (EA) were more prominent in Langerhans islets [glucose-6-phosphatase, glucose-6-phosphate dehydrogenase (DH), isocitrate DH, glycerol-3-phosphate DH, NADPH DH], others were strongly marked in acini and ducts (alkaline phosphatase, beta-glucuronidase, acid esterase aryl-sulfatase). Histochemical and enzyme abno… Show more

“…Many morphological studies have dealt with the pancreas of various animals after duct obstruction either by ligation or injection of foreign materials such as microspheres or high-viscosity solutions into the duct (EDLUND et al, 1962;SATAKE et al, 1983;EKBERG et al, 1988;KLEMPNAUER et al, 1990;CATALA et al, 1990). The findings in those studies, however, depended on limited fields of thin sections, lacking in extensive and three-dimensional information concerning the whole organ.…”

“…lc). The basal plasma glucagon of 158 _ 15 ng/1 in the C group remained relatively stable over 450 days. It ranges from 120pg/ml in humans I181 and 180pg/ml in dogs I411 to 323 pg/ml and 331pg/ml in Wistar and GK rats, respectively (1).…”

“…Another method of surgically inducing DM was described in rabbits as early as 1946 when Walpole and Innes performed a pancreatic duct ligation, which resulted in morphological changes of the islets and a slight rise in the fasting blood glucose level. Later, this model was investigated in more detail and assigned to T2DM models [21,22].…”
