Cell Proliferation and Cytotoxicity Assays

Abstract: Cell viability is defined as the number of healthy cells in a sample and proliferation of cells is a vital indicator for understanding the mechanisms in action of certain genes, proteins and pathways involved cell survival or death after exposing to toxic agents. Generally, methods used to determine viability are also common for the detection of cell proliferation. Cell cytotoxicity and proliferation assays are generally used for drug screening to detect whether the test molecules have effects on cell prolifer… Show more

“…Following 24 h culture period, macrophages were washed with PBS, detached via gentle scraping and finally collected by centrifugation at 240 x g, 4˚C, for 5 min. Cell viability was evaluated by the trypan blue exclusion method (24) and was >80% in all experiments. Macrophages were seeded at an initial density of 7x10 5 viable cells in 24-well plates and allowed to adhere for 24 h. Macrophages were then cultured in growth medium supplemented with polymyxin B (25 µg/ml, Sigma-Aldrich; Merck KGaA) and then either left untreated or treated with 25 µg/ml of mCRP, pCRP, oxLDL (medium oxidized low density lipoprotein, Kalen Biomedical, LLC, Germantown, MD, USA) alone or in combination.…”

“…After treatment with PLLA extract or Cur in 12 hr, 24 hr, and 48 hr, respectively, cell viability was assessed by CCK‐8 according to the manufacturer's instructions. CCK‐8 contains the tetrazolium salt WST‐8 (Semisch & Hartwig, ) and is markedly more convenient than MTT for cell viability assessment (Adan, Kiraz, & Baran, ). Absorbance was measured at 450 nm on a microplate reader (Multiskan FC, Thermo Scientific).…”
