Cell surface antigens of human bladder cancer defined by mouse monoclonal antibodies.

Abstract: Mouse monoclonal antibodies were obtained by immunization with cultured human bladder cancer or lysates of bladder papilloma. They identify 11 distinct antigenic systems as defined by serological analysis of cultured cells and studies of antigen distribution in normal and neoplastic tissues. The most restricted of these antigens, OmS, defines a subset of bladder tumors. Om5 is not detected in normal bladder urothelium or in any other normal or malignant tissue. T101 and JP165 are also subset markers for bladde… Show more

“…Monoclonal antibodies reactive with TCC associated antigens have recently been reported by other groups (Fradet et al, 1984;Mazuko et al, 1984;Messing et al, 1984;Grossman, 1983). However, comparison of these findings with our own data suggests that none of these antibodies is likely to be identical to the ones described herein.…”

“…Recent reports on different tumour antigens associated with urinary bladder cancer (Fradet et al, 1984;Mazuko et al, 1984;Koho et al, 1984;Messing et al, 1984;Grossman, 1983) suggest the existence of a complex group of TAAs similar to what has been found for melanomas (for review see Hellstr6m et al, 1985). In this study we describe the production and specificity patterns of two new monoclonal antibodies extending our earlier analysis of the heterogeneity of TAAs in human bladder carcinoma.…”

“…Recombinant humanized ONS-M21 Ab has already been successfully produced (Ohtomo et al, 1995), and therefore hONS-M21 Ab is the only Ab against gliomas whose target molecule has been identified. The expression of integrin α3 in the normal brain has been reported to be very low or even undetectable (Fradet et al, 1984;McGeer et al, 1990;Paulus et al, 1993), and ONS-M21 MAb is not reactive to either fetal or adult brain tissues (Moriuchi et al, 1993). A significant amount of ONS-M21 MAb attached to the cell surface was rapidly internalized into the cytoplasm, and the A-chain of ricin-conjugated ONS-M21 MAb displayed glioma specific cell toxicity in vitro (Shimizu et al, 1998).…”

“…The human trophoblast cell-surface antigen, TROP2 (also termed GA733-1), is a transmembrane glycoprotein found to be expressed at high levels by various types of human carcinomas (Fradet et al, 1984;Stein et al, 1993a;Nakashima et al, 2004). In contrast, normal epithelial tissues show little or no TROP2 expression (Stein et al, 1993a;Zhang et al, 1997).…”
