Cellular immunotherapy for hematological malignancy: recent progress and future perspectives

Abstract: Advancements in the field of cellular immunotherapy have accelerated in recent years and have changed the treatment landscape for a variety of hematologic malignancies. Cellular immunotherapy strategies exploit the patient's immune system to kill cancer cells. The successful use of CD19 chimeric antigen receptor (CAR) T-cells in treating B-cell malignancies is the paradigm of this revolution, and numerous ongoing studies are investigating and extending this approach to other malignancies. However, resistance t… Show more

“…In non-small cell lung cancer, higher expression of GSDMD is associated with giant tumors and more advanced cancer stages. Pyroptosis is also associated with many adverse effects of cancer therapy, such as cytokine release syndrome in CAR T cell therapy 39 , 118 or chemotherapy drug damage to normal tissues in chemotherapy 35 . Additionally, pyroptosis induces the secretion of proinflammatory cytokines, and chronic inflammation has traditionally been known to promote tumorigenesis.…”

“…Chimeric antigen receptor (CAR)-T cells have a major role in hematologic malignancies. With the design of different CAR structures, applications of CAR-T therapies have been expanded from B-cell to other hematologic malignancies 3 ; however, cytokine release syndrome (CRS) in patients undergoing autologous CAR-T-cell therapy and GVHD to allogenic CAR-T cells limit for further application 4 , 5 .…”
