Cellular uptake studies of two hexanuclear, carboxylate bridged, zinc ring structures using fluorescence microscopy

Abstract: Two hexanuclear zinc complexes have been structurally characterized, and evaluation against several cells showed selective toxicity. Cellular uptake revealed a non-specific process, resulting in accumulation within the cell cytoplasm.

“…The reaction of this ligand with a cobalt(II) pivalate (trimethyl acetate) precursor in the presence of a base, triethylamine, in acetonitrile afforded a new pentanuclear mixed‐valent cobalt cluster of formula [Co II 3 Co III 2 (OH) 2 (piv) 6 (L) 2 (H 2 O) 4 ] (piv = trimethylacetate). One hexanuclear {Zn 6 } compound10 and a couple of dinuclear {Cu 2 } examples11 are the only reported 3d metal complexes bearing this H 2 L ligand, enhancing the relevance of this new {Co 5 } polynuclear compound.…”

“…Quantum yields (φ F ) of the probes 1a-m were measured using quinine sulfate in 1 N H 2 SO 4 as the standard. [23] The highest φ F was observed for 1f, and the lowest one was observed for 1i ( Figure 3). The emission bands of compound 1i (λ em =402 and 360 nm) are due to the quinazolin-4(3H)-one moiety and 2-hydroxy-3-methoxyphenyl groups, respectively, the latter of which as a chromophore shows a significant absorption at 334 nm (Figure 3, spectrum 1i).…”

“…The calculated absorption and emission bands are in excellent agreement with experimentally observed values. Quantum yields (φ F ) of the probes 1a‐m were measured using quinine sulfate in 1 N H 2 SO 4 as the standard . The highest φ F was observed for 1f , and the lowest one was observed for 1i (Figure ).…”

“…This is supported by similar experiments conducted by Redshaw et al to see the cellular localisation of fluorescent zinc complexes using DAPI as a counterstain. 24 These drug localisation experiments only gave information about whether the drug is present in the nucleus or not. By measuring the amount of drug taken up by the cells after treatment for particular time will give quantitative information about the rate of drug uptake in the cells.…”

“…DS26 is a self-complementary DNA sequence 5′-CA2TCG2ATCGA2T2CGATC2GAT2G-3′] which form a duplex DNA. 24 The gradual decrease in the fluorescence arising from displacement of thiazole orange from quadruplex and duplex DNA is used to profile the binding upon gradual increase of the ligand concentration.…”
