Ceramide Acyl Chain Length and Its Relevance to Intracellular Lipid Regulation

Abstract: Ceramides are a class of sphingolipids which are implicated in skin disorders, obesity, and other metabolic diseases. As a class with pleiotropic effects, recent efforts have centred on discerning specific ceramide species and their effects on atopic dermatitis, obesity, type 2 diabetes, and cardiovascular diseases. This delineation has allowed the identification of disease biomarkers, with long acyl chain ceramides such as C16- and C18-ceramides linked to metabolic dysfunction and cardiac function decline, wh… Show more

“…Ceramides of 24- or 16-carbon fatty acyl chain length are known to differentially impact cell fate (for reviews see [ 65 ]). Evidence suggests that ceramides with 24-carbon fatty acid acyl chain lengths are pro-survival lipids as they have cytoprotective effects, promote cellular growth, and have anti-apoptotic properties [ 66 , 67 , 68 ].…”

“…It is known that the ceramide content remains low in non-dividing cells, but can increase due to exposure to stress conditions, such as serum starvation, chemical compound exposure, or oxidative stress conditions [ 48 ]. These sphingolipids, composed of sphingosine group and fatty acid, are primarily produced by de novo synthesis [ 49 ], the salvage pathway, or sphingomyelinase pathways [ 50 ]. We are able to detect an increase in ceramides in Paraquat-treated membranes, as all adducts increase their presence, whereas, with α-tocopherol pre-treatment, the unsaturated ceramides decrease with respect to their values in Paraquat treatment, or remain absent.…”
