Challenges in the Development, Licensure, and Use of Combination Vaccines

Abstract: Before substantial public health benefits associated with use of combination vaccines can be realized, a variety of challenges must be addressed. In February 2000, the National Vaccine Program Office convened the International Symposium on Combination Vaccines to explore solutions for barriers to development, licensure, and use of safe and effective combination vaccines. The symposium focused on the following questions: (1) What immunologic standards should be used to evaluate new combination vaccines? (2) How… Show more

“…Also, inactivated poliovirus vaccine combined with diphtheria and TT and whole cell pertussis vaccine diminished antibody response to pertussis vaccine [ 30 ]. Conversely, whole cell pertussis vaccine combined with diphtheria toxoid boosted the immune response to individual components [ 31 , 32 ]. In our model, although the combination of OVA plus TT decreased TT-specific antibodies indicating antigenic completion, the addition of LPS potentiates IgG1 and IgG2a anti-TT antibodies while decreasing IgE antibodies revealing that LPS boost the production of anti-infectious antibodies.…”

“…A combined vaccine is that one consists of 2 or more separate immunogens physically combined into a single product [16]. Several benefits of a combined vaccine must be fulfilled as aiming to prevent multiple diseases caused by different organisms [17] and also reducing the number of injections and therefore the degree of pain and side effect of repeated vaccination.…”

“…Such vaccines, that combine antigens from two or more different vaccines, are often submitted for licensure on the basis of non-inferiority trials that demonstrate safety and immunogenicity profiles similar to component vaccines administered separately [15]. Combination vaccines are often proposed as a future solution that may help overcome the many ethical and regulatory issues in developing second-generation vaccines: a new antigen could be included in combination with the antigen(s) in the licensed vaccine (for example, RTS,S plus antigens).…”
