Changes in long non-coding RNA transcriptomic profiles after ischemia-reperfusion injury in rat spinal cord

Abstract: With the aim of exploring expression profiles and biological functions of long non-coding RNA (lncRNA) and mRNAs after spinal cord ischemia-reperfusion injury (SCII), differentially expressed lncRNAs (DElncRNAs) and mRNAs (DEmRNAs) in rat spinal cords were identified following SCII through high-throughput RNA sequencing. In total, 1,455 lncRNAs and 6,707 mRNAs were observed to be differentially expressed (—Fold Change— ≥ 2 and P < 0.05) after SCII, including 761 up-regulated and 694 down-regulated lncRNAs, … Show more

“…Previous studies have demonstrated that miRNAs and lncRNAs are potentially new regulators in prevention and treatment of SCII and their roles in regulating target genes might be crucial in the pathological mechanism of SCII ( Hu et al, 2013 ; Li J. A. et al, 2016 ; Zhou et al, 2020c ). In rat models of SCII, miR-125b mimic was found to protect against SCII via reducing aberrant p53 network activation-induced apoptosis and neuroinflammation through the downregulation of TP53INP1 ( Li et al, 2018 ).…”

“…The miRNAs have been shown to regulate genes associated with the pathological mechanism of SCII (13,14,29). For example, miR-30c protects PC12 cells against oxygen-glucose deprivation (OGD)-caused apoptosis and inflammatory response and inhibits the development of SCII by modulating the expression of SIRT1 (30).…”

“…Recent evidence has indicated that many lncRNAs are differentially expressed in SCIRI and may act as a significant part of the pathophysiology of SCIRI. Zhou et al analyzed the expression pattern of lncRNAs in the SCIRI rat model using high-throughput RNA sequencing, suggesting that 6,707 mRNAs and 1,455 lncRNAs were differentially expressed, including 761 lncRNAs with increased expression and 694 with decreased expression ( 83 ). Furthermore, another study profiled the expression pattern of lncRNAs using a similar method and indicated that 7,980 lncRNAs (234 upregulated and 7,746 downregulated) and 3,428 mRNAs were differentially expressed ( 84 ).…”

A bioinformatics analysis showed that those differentially expressed lncRNAs and mRNAs were associated with many biological pathways and processes, which might be crucial to provide new insight into the identification of potential therapeutic targets for SCIRI ( 83 ).…”
