Changes of MicroRNA-1, -15b and -21 Levels in Irradiated Rat Hearts After Treatment With Potentially Radioprotective Drugs

Abstract: The aim of this study was to measure expression levels of microRNAs (miRNAs) (miRNA-1, -15b and -21) in the rat myocardium after a single dose of ionizing radiation (6-7 Gy/min, total 25 Gy). The rats were treated with selected drugs (Atorvastatin, acetylsalicylic acid (ASA), Tadalafil, Enbrel) for six weeks after irradiation. MiRNAs levels were measured by RT-qPCR. Irradiation down-regulated miRNA-1 in irradiated hearts. In Tadalafil- and Atorvastatin-treated groups, miRNA-1 expression levels were further dec… Show more

“…Among the miRs connected with radiation-induced heart disease are also miR-1 and miR-21. Kura et al [103] reported the down-regulation of miR-1 and the up-regulation of miR-21 in the rat myocardium six weeks after single irradiation of the mediastinum area with a total dose of 25 Gy. These results were confirmed in their next study in 2019 when RT-qPCR proved the down-regulation of miR-1 and the up-regulation of miR-21 in rats' left ventricle after 10 Gy of irradiation in the mediastinal area.…”

“…In irradiated rats, miR-1 was found downregulated in the heart [87,139]. miR-1 plays a pivotal role in electrical coupling and direct cardiac cell to cell communication to ensure heart function, since it targets intercellular Cx43 channels [140].…”

“…The expression of some miRNAs is highly tissue-specific, e.g. miR-1 and -133 (cardiac and skeletal muscle), miR-122 (liver), and miR-124a (brain) (Lagos-Quintana et al 2002). Among others, kidney-specific miRNAs (miR-192, -194, -204, and -215) Of the cardiac-specific miRNAs, miR-1 acts as a regulator of differentiation and proliferation during cardiogenesis and as a regulator of cardiomyocyte growth in the adult heart.…”

“…Of the possible epigenetic mechanisms that lead to disease development, in addition to DNA methylation (almost exclusively cytosine methylation within the sequences with a high proportion of CG repeats) and histone modification (methylation, acetylation, phosphorylation, ubiquitination) more and more attention is now being focused on the analysis of regulatory RNA molecules (Udali et al 2013, Kuneš et al 2015, Kura et al 2016.…”
