Characterisation of micromonosporae from aquatic environments using molecular taxonomic methods

Abstract: Large numbers of strains assigned to the genus Micromonospora on the basis of typical colonial and pigmentation features were isolated from diverse aquatic sediments using a standard selective isolation procedure. Two hundred and six isolates and eight representatives of the genus Micromonospora were assigned to 24 multimembered groups based on a numerical analysis of banding patterns generated using BOX and ERIC primers. Representatives of multimembered groups encompassing isolated micromonosporae were the su… Show more

“…There is indeed strong evidence suggesting that Micromonospora type strains, genomically defined by lower than about 70% DNA-DNA reassociation values, are delineated by threshold values of almost 99.3% 16S rRNA gene sequence similarities (Kasai et al, 2000;Thawai et al, 2007;Trujillo et al, 2006Trujillo et al, , 2007Jongrungruangchok et al, 2008;Maldonado et al, 2008). In this study, many of the strains sequenced were found in this range.…”

“…BOX-PCR fingerprinting has been shown to be a useful tool to discriminate highly related strains and has been applied to study the genetic diversity of different bacterial taxa including Micromonospora (Lanoot et al, 2004;Maldonado et al, 2008;Torres et al, 2008). To get more insight into the genetic diversity of the Micromonospora strains isolated, they were analysed by BOX-PCR fingerprinting.…”

“…Marine sediments have proven a profi table habitat for the isolation of high numbers of Micromonosporaceae (Bredholt et al, 2008;Maldonado et al, 2008;Prieto-Davó et al, 2008). Our isolate, Micromonospora strain DB620, originated from the German Wadden Sea, a unique ecosystem characterized by large areas of tidal fl ats.…”
