Characteristics and functions of lipid droplets and associated proteins in enterocytes

Abstract: Cytosolic lipid droplets (LDs) are observed in enterocytes of jejunum during lipid absorption. One important function of the intestine is to secrete chylomicrons, which provide dietary lipids throughout the body, from digested lipids in meals. The current hypothesis is that cytosolic LDs in enterocytes constitute a transient pool of stored lipids that provides lipids during interprandial period while lowering chylomicron production during the post-prandial phase. This smoothens the magnitude of peaks of hypert… Show more

“…In addition, specific isoforms of enzymes that activate FAs to form acyl-CoAs for incorporation into TAG (ACSL1, ACSL3, and ACSL4) have been identified on CLDs in several cell types (reviewed in [118]). In enterocytes, TAG synthesis enzymes that have been identified on CLDs are ACSL3, ACSL4, ACSL5, GPAT3 and MGAT2 in mice and humans (reviewed in [45, 119]). Of these, ACSL3 [120], ACSL5 [97], and GPAT3 [45] have been validated by confocal or electron microscopy to be present on CLDs in enterocytes.…”

“…The proteomes of isolated CLDs derived from Caco-2 cells and mouse enterocytes have been analyzed using a tandem mass spectrometery approach [97, 120, 125] and have been previously reviewed [119]. These studies identified both known and novel CLD associated proteins.…”

“…DHSRS3 is localized in the endoplasmic reticulum (ER) and lipid droplets, both of which are storage vesicles containing lipid, such as cholesterol and retinol esters [48]. Additionally, overexpression of DHRS3 in U2OS cells increases lipid droplet accumulation (Figure 1B) [26].…”

“…The bulk of dietary PUFAs is in the form of neutral fat or triglyceride and is well absorbed in the small intestine via emulsification, hydrolysis, and micelle formation (15). Following lipoprotein transport, PUFAs are incorporated into cell membrane glycerophospholipids by a remodeling pathway (Lands' cycle) to generate membrane asymmetry and diversity (194).…”
