Characteristics ofBorrelia hermsiiinfection in human hematopoietic stem cell-engrafted mice mirror those of human relapsing fever

Abstract: Rodents are natural reservoirs for a variety of species of Borrelia that cause relapsing fever (RF) in humans. The murine model of this disease recapitulates many of the clinical manifestations of the human disease and has revealed that T cell-independent antibody responses are required to resolve the bacteremic episodes. However, it is not clear whether such protective humoral responses are mounted in humans. We examined Borrelia hermsii infection in human hemat… Show more

“…IgM appeared to play a major role in B. miyamotoi clearance in wild-type mice, as its rise coincided with B. miyamotoi clearance, and passive transfer of 5-day immune sera to SCID mice cleared spirochetemia. In other relapsing fever spirochetes, T-cell independent IgM has a major role in early clearance, facilitated by B1b cells and splenic marginal zone B cells (21, 46, 48, 56). IgM specific for Vlp7 has been demonstrated to directly neutralize Vlp7-expressing B. hermsii in vitro and in vivo upon passive immunization.…”

“…Anti- B. hermsii responses were diminished and persistent bacteremia recurred upon administration of anti-human CD20 antibodyVuyyuru et al (2011)DENVNOD/ scid engrafted with human fetal thymus and liver tissue under the kidney capsule and intravenously injected with CD34 + human FL cells to create huBLT mice. Mice were intravenously infected with DENV-2Intravenous inoculation of DENV-2 resulted in sustained viremia and infection of leukocytes in lymphoid and non-lymphoid organs.…”

“…Since then several independent groups have evaluated this small circulating B-cell population in healthy volunteers and in clinical conditions of common variable immunodeficiency, multiple sclerosis, bone marrow transplantation, and lupus, and the proposed CD20 + CD27 + CD43 + B-1 cells have been shown to respond to pneumococcal vaccination and to be associated with protection against Borrelia infection (28-34). …”
